Position Summary:

Poseida is seeking an outstanding candidate to join our Gene Therapy team. The Genotoxicity Research Scientist will play a key role investigating the effects of gene editing and transposon integration using our platform technologies (Cas-CLOVER, piggyBac transposon, and site-specific piggyBac transposon) for therapeutic applications for gene therapies.


Responsibilities

These may include but are not limited to:

  • Apply your strong background in molecular biology, genetics, transposons and/or site-specific nucleases to characterize genetic manipulations generated using the piggyBac and Cas-CLOVER platforms.
  • Develop new and optimize existing customized NGS analysis tools for internal R&D use and in support of regulatory filings.
  • Generate extensive reports, for documentation of gene therapy/editing genotoxicity, suitable for regulatory filings.
  • Work in a highly collaborative, cross-functional team environment to perform experiments in the lab, analyze results, and communicate findings in team meetings.
  • Keep current on trends, new products and technologies in gene editing and knock-ins.
Requirements

Requirements, Knowledge, Skills, and Abilities:

  • Ph.D. in a scientific discipline (e.g., molecular biology, cell biology, or related field) with 0-2 plus years of hands-on experience with gene editing, site-specific nucleases (especially RNA-guided nucleases), transposons, or other vectors used in vivo or in vitro, is required.
  • A strong background in standard molecular biology (such as PCR, qRT-PCR, ddPCR, DNA/RNA extraction and molecular cloning) is required.
  • Experience with mammalian cell culture is required, and any experience with the culture and genetic manipulation of primary human hepatocytes, organoids, T-cells, or other cells, is a plus.
  • Prior experience with viral or non-viral gene delivery technologies (transposon, episomal/plasmid, minicircle, or nanoparticles) is required.
  • Intimate knowledge or direct experience with sequencing library preparation (e.g. Illumina, Nanopore, or PacBio platforms) is strongly preferred.
  • Previous experience using methods to investigate/query off-target nuclease-mediated editing, such as GUIDE-seq, ONE-seq, Circle-seq, or amplicon-seq is preferred, but not required.
  • Previous experience using methods to investigate/query transposon or viral vector integration sites, such as LM-PCR or LAM-PCR, is preferred.
  • Proficiency with multi-parameter flow cytometry, cell sorting, and FACS analysis software is a plus.
  • Experience R, Python, shell scripting, and bioinformatics analysis, is a plus.
  • Must possess excellent oral and written communication skills.


Research Scientist (Genotoxicity) Pay Rate: $100,000 to $118,000 annually

The final pay offered to a successful candidate will be dependent on several factors that may include but are not limited to the type and length of experience within the job, type, and length of experience with the industry, education, geographic location, etc.
